Conservation genomics of Perdita meconis: An exemplary study for rare desert specialist bees

Perdita meconis, the Mojave poppy bee, is a rare desert specialist endemic to the eastern Mojave Desert. It is currently under review for listing under the Endangered Species Act (ESA), as its range and abundance have potentially declined due to habitat loss and the disappearance of its host plants. While recent research has focused on presence-absence surveys and the potential replacement of its pollination niche by other species, the genetic health of P. meconis is completely unknown.
This study uses whole genome re-sequencing data from recently collected specimens and a newly published reference genome to investigate population genomics across its range. Key objectives include assessing overall genetic diversity, identifying the number of genetically distinct populations, characterizing levels of gene flow, and detecting potential signs of inbreeding. Given the species’ small body size, likely limited dispersal ability, and fragmented habitat, we investigate an isolation-by-distance pattern among populations. To better understand historical population dynamics, we also explore the use of pairwise sequentially Markovian coalescent (PSMC) modeling to reconstruct past changes in effective population size.
As one of few population genomic studies on desert solitary bees, this work will provide a reference point for solitary bees more generally. The findings will contribute to conservation planning and ESA listing decisions, while also offering broader insight into the biology and dispersal ecology of this understudied pollinator.
